Add this to your prefs.js file in \(NS dir)\Users\(profile directory):

user_pref("os2.mail.sound", "E:\\MMOS2\\SOUNDS\\APPLAUSE.WAV");

Note the double slashes.  You can, of course, use a .wav of your
choosing.
