  I assume you have rebooted (in the case SP firewall turned off by
policy).   Then

   netstat -an
   to verify that port 25 is listening.

From the server:
  telnet 127.0.0.1 25
  to verify that you can connect locally.   Then just back up step by step
to find where the block occurs.
