You didn't mention what OS that your running.
with OS/2 the line in the config.sys is, mine anyhow:

Under dos locking the port usually involves using a fossil driver or the mode 
command
I would list that but it has been over 4 years since I used dos that I 
forgotten that one...
using a fossil driver in dos ie X00,
device=X00.sys com2:57600    something like that anyhow...
Other people using other OS's will probably be able to help...
