There are several possible causes for peaks in network traffic:
  - backup via the LAN on regular times (typically once per day)
  - domain controller replication (by default, this happens more often
than just once per day). Ditto for DNS and WINS servers
  - refreshing virus signature files on all servers and workstations
(typically once per day)
  - manual sync of all domain controllers, e.g. after inputting a batch
of new users, changing access restrictions or resetting some passwords.
This can also happen at fixed times when the person in charge of user
management is instructed to always do a sync at the end of each working day
  - etc etc

All these can (but should not!) cause the phenomena in question. Take your pick.

Geo's hint to use a sniffer can narrow down the wealth of possibilities to
just a few. But without a knowledgable network guy, this won't work.
