All Microsoft products must boot from C:\ but OS/2 is not restricted thus and, for example, my own system boots Dos 6.3/Windows from FAT C:\ and Blue Spine Warp 3 from HPFS F:\, using Boot Manager in a 1meg primary partition (which is not, however, allocated a drive-table letter).
