I've booted Linux Ubuntu from an external hard drive without any problem whatsoever. Likewise, OS X can be booted from an external hard drive. Although running any OS from an external hard drive is slow even with Firewire 800 technology. Maybe it will be more doable now that Apple has included the Thunderbolt port on all new Macs. There are still no Thunderbolt devices available yet but are forthcoming.
