Samples from the Command Reference, located in your help folder:

To install a serial device on COM4 for ISA/EISA bus computers, type
the following in the CONFIG.SYS file: 

DEVICE=C:\OS2\BOOT\COM.SYS (4,2e8,10)


To install serial devices on COM3 and COM4 for ISA/EISA bus
computers, type the following in the CONFIG.SYS file: 

DEVICE=C:\OS2\BOOT\COM.SYS (3,3e8,5) (4,2e8,9)

The first parameter is com port, the next is the standard i/o port
for said com port, and the last is the IRQ line that the device is
set to.  This refers to a device where you set the IRQ with
switches or vendor software.

This will work on pci also if you can set certain slots to legacy
mode.
