From what I have read and experienced:
 It's more comfortable without.
The stuff the spandex is made out of is made so that it takes away moisture
and dries your skin/sweat, making everything feel pretty good. The seams are
cut in a certain pattern as to not affect your rear end or groin section,
there's padding in most of them, etc.
If you throw in a pair of.... Hanes.. or whatever, they get bunched up, take
away from ALL of the moisture absorbing properties of the spandex, and alter
the effects of the padding that the shorts have, etc.
I, at first, always wore underwear with them, cause I never even thought of
*Not* wearing..
Then I think it was bicycling magazine, that reported that as being one of
the most common, "amateur" mistakes, that no-one talks about so how would
anyone know, right?
So, I tried it, and it feels better and works fine..
